Indications:
Beklo is indicated for:

  • Relief of muscle spasticity due to multiple sclerosis

  • Muscle spasticity resulting from spinal cord injury or disease

  • Cerebral palsy, stroke, or other neurological disorders with spasticity

Pharmacology:
Baclofen is a centrally acting skeletal muscle relaxant. It is a gamma-aminobutyric acid (GABA) derivative that acts on GABA-B receptors in the spinal cord, reducing the release of excitatory neurotransmitters and inhibiting reflexes responsible for muscle spasticity. This results in muscle relaxation and relief from stiffness and spasms.

Dosage & Administration:

  • Initial dose: 5 mg orally three times daily

  • May be increased by 5 mg per dose every 3 days as needed

  • Maintenance dose: Usually 40–80 mg/day in divided doses

  • Take with food to reduce gastrointestinal discomfort

  • Do not stop abruptly — taper gradually to prevent withdrawal symptoms

Interaction:

  • Enhanced CNS depression with alcohol, benzodiazepines, opioids, antihistamines

  • May enhance the effects of other muscle relaxants

  • Caution with tricyclic antidepressants (may potentiate muscle weakness)

Contraindications:

  • Hypersensitivity to Baclofen

  • Active peptic ulcer (relative)

  • Use cautiously in patients with epilepsy or renal impairment

Side Effects:

  • Common: Drowsiness, dizziness, weakness, nausea, fatigue

  • Less common: Headache, hypotension, confusion, constipation

  • Rare: Seizures, hallucinations (particularly with abrupt withdrawal), respiratory depression

Pregnancy & Lactation:
Use in pregnancy only if clearly needed; safety not established. Baclofen is excreted in breast milk; use with caution during lactation under physician supervision.

Precautions & Warnings:

  • Avoid activities requiring mental alertness (e.g., driving) until drug effects are known

  • Use cautiously in elderly and renal-impaired patients

  • Do not discontinue suddenly to avoid hallucinations, seizures, or rebound spasticity

  • Monitor renal function in long-term use
