Indications:
Ropitor is indicated for:
Hyperlipidemia and mixed dyslipidemia
Primary and secondary prevention of cardiovascular events
Homozygous familial hypercholesterolemia (HoFH)
Slowing progression of atherosclerosis
Pharmacology:
Rosuvastatin is a selective and competitive inhibitor of HMG-CoA reductase, the rate-limiting enzyme in cholesterol biosynthesis. It increases the number of LDL receptors on hepatocyte surfaces, enhancing LDL uptake and breakdown, and inhibits hepatic synthesis of VLDL, lowering overall LDL and triglyceride levels while increasing HDL.
Dosage & Administration:
Usual starting dose: 5–10 mg once daily
Can be taken with or without food, preferably at the same time every day
Dose adjusted based on lipid goals and patient tolerance
Regular lipid profile monitoring recommended
Interaction:
Increased risk of myopathy with cyclosporine, gemfibrozil, or other fibrates
Use with caution with antacids (take Ropitor 2 hours before or 4 hours after antacids)
May increase INR in patients taking warfarin
Contraindications:
Hypersensitivity to Rosuvastatin or any component
Active liver disease or unexplained persistent elevations in hepatic transaminases
Pregnancy and lactation (contraindicated)
Side Effects:
Common: Headache, muscle pain, abdominal pain, nausea
Less common: Liver enzyme elevation, weakness
Serious (rare): Rhabdomyolysis, myopathy, kidney dysfunction
Pregnancy & Lactation:
Contraindicated in pregnancy and breastfeeding due to potential harm to fetus or infant. Use effective contraception during therapy.
Precautions & Warnings:
Monitor liver enzymes before and during treatment
Discontinue if symptoms of myopathy or rhabdomyolysis occur
Use lower doses in Asian patients or those with renal impairment
