Indications:
AMITIP 10MG is indicated for the treatment of depression, especially endogenous depression. It is also used in the management of anxiety disorders, migraine prophylaxis, and certain types of chronic pain such as neuropathic pain and fibromyalgia.
Pharmacology:
Amitriptyline is a tricyclic antidepressant. It works by inhibiting the reuptake of norepinephrine and serotonin at the nerve endings in the central nervous system, thereby increasing the availability of these neurotransmitters and improving mood.
Dosage:
Adults: 25–75 mg daily in divided doses or a single dose at bedtime. Dosage may be gradually increased if necessary.
Elderly: Lower initial doses (10–25 mg/day) are recommended.
Children: Use is not recommended unless prescribed by a specialist.
Administration:
Should be taken with or after food to minimize gastrointestinal discomfort. Preferably taken at night due to sedative effects.
Interaction:
May interact with MAO inhibitors, SSRIs, anticholinergic agents, antihypertensive drugs, alcohol, and other CNS depressants.
Contraindications:
Contraindicated in patients with recent myocardial infarction, arrhythmias, or hypersensitivity to amitriptyline or other tricyclic antidepressants.
Side Effects:
Common side effects include dry mouth, drowsiness, dizziness, weight gain, blurred vision, and constipation. Serious side effects may include arrhythmias, seizures, and urinary retention.
Pregnancy & Lactation:
Should be used in pregnancy only if clearly needed. Amitriptyline is excreted in breast milk; caution is advised during lactation.
Precautions & Warnings:
Caution in patients with cardiovascular disease, glaucoma, urinary retention, epilepsy, and liver impairment. Abrupt discontinuation should be avoided.
