Indications:
AMITIP 25MG is indicated for the treatment of major depressive disorder, anxiety disorders, migraine prevention, and chronic pain syndromes such as neuropathic pain and fibromyalgia.
Pharmacology:
Amitriptyline is a tricyclic antidepressant (TCA). It acts primarily by blocking the reuptake of norepinephrine and serotonin in the brain, which enhances neurotransmitter activity and improves mood and pain perception.
Dosage:
Adults: Initially 25 mg at bedtime, which may be gradually increased up to 150 mg daily in divided doses or a single dose.
Elderly or debilitated patients: Start with 10–25 mg daily and increase cautiously.
Children: Not generally recommended unless prescribed by a psychiatrist.
Administration:
Should be taken preferably at night with or after meals to reduce gastric irritation and maximize sedative effects.
Interaction:
May interact with alcohol, MAO inhibitors, SSRIs, anticholinergics, sedatives, antihypertensives, and other CNS-active drugs.
Contraindications:
Hypersensitivity to amitriptyline, recent myocardial infarction, arrhythmias, and co-administration with MAO inhibitors are contraindicated.
Side Effects:
Common side effects include dry mouth, sedation, blurred vision, weight gain, constipation, and dizziness. Rare but serious effects include cardiac arrhythmia, seizures, and suicidal thoughts.
Pregnancy & Lactation:
Use only if potential benefit justifies potential risk. Amitriptyline may pass into breast milk; caution should be exercised during breastfeeding.
Precautions & Warnings:
Use with caution in patients with heart disease, epilepsy, urinary retention, liver dysfunction, glaucoma, and in elderly patients. Do not stop the drug abruptly.
