Indications:
UNIGERD is indicated for the relief of nausea and vomiting of various causes, symptoms of bloating, early satiety, and upper abdominal discomfort. It is also used in gastroparesis and reflux-related disorders.
Pharmacology:
Domperidone is a dopamine D2 receptor antagonist with prokinetic and antiemetic properties. It increases lower esophageal sphincter pressure, enhances gastric emptying, and coordinates gastric contractions without affecting gastric acid secretion.
Dosage:
Adults: 10 mg up to 3 times daily, 15–30 minutes before meals and, if necessary, before bedtime. Maximum dose usually should not exceed 30 mg per day.
Administration:
Take orally before meals. Do not take with grapefruit juice or high-fat meals as it may delay absorption.
Interaction:
May interact with ketoconazole, erythromycin, fluconazole, and some antidepressants. Avoid use with QT-prolonging drugs or in patients with existing heart conditions.
Contraindications:
Hypersensitivity to Domperidone, prolactin-releasing pituitary tumor (prolactinoma), gastrointestinal bleeding, perforation or mechanical obstruction, and moderate to severe liver dysfunction.
Side Effects:
Common: Dry mouth, abdominal cramps, dizziness.
Rare: Irregular heartbeat, galactorrhea, gynecomastia, QT prolongation (especially in elderly patients).
Pregnancy & Lactation:
Not recommended during pregnancy unless clearly necessary. Domperidone is excreted in breast milk in small amounts; consult a doctor before use during lactation.
Precautions & Warnings:
Use with caution in patients with cardiac disease, electrolyte imbalance, or kidney/liver impairment. Prolonged use not advised without physician supervision.
