Indications:
Comet® is indicated for:
Management of type 2 diabetes mellitus, especially in overweight patients
Improving glycemic control when diet and exercise alone are insufficient
May be used alone or in combination with other antidiabetic agents
Pharmacology:
Metformin decreases hepatic glucose production, decreases intestinal glucose absorption, and improves insulin sensitivity by increasing peripheral glucose uptake and utilization, resulting in better glycemic control without causing hypoglycemia.
Dosage & Administration:
Adults: Usually start with 500 mg once or twice daily, may increase up to 2000 mg/day based on tolerance and response.
Take with meals to reduce gastrointestinal side effects.
Interaction:
May interact with cimetidine, iodinated contrast agents, and other drugs affecting renal function.
Alcohol increases risk of lactic acidosis.
Contraindications:
Renal impairment or dysfunction
Metabolic acidosis including diabetic ketoacidosis
Hypersensitivity to Metformin
Side Effects:
Common: gastrointestinal disturbances (nausea, vomiting, diarrhea)
Rare but serious: lactic acidosis
Vitamin B12 deficiency with long-term use
Pregnancy & Lactation:
Use during pregnancy only if clearly needed. Consult doctor before use during breastfeeding.
Precautions & Warnings:
Monitor renal function periodically
Avoid alcohol consumption
Use caution in elderly and in patients with hepatic impairment
