Indications:
Tryptin is indicated for the treatment of:

Major depressive disorder
Anxiety disorders
Neuropathic pain
Migraine prophylaxis
Nocturnal enuresis in children (bedwetting)
Chronic tension-type headache
Fibromyalgia

Pharmacology:
Amitriptyline is a tricyclic antidepressant (TCA). It inhibits the reuptake of norepinephrine and serotonin, increasing their levels in the central nervous system. It also has sedative and anticholinergic effects.

Dosage & Administration:

Depression/Anxiety: Usually 25–75 mg daily in divided doses or at bedtime; dose can be increased gradually
Neuropathic pain: Often started at 10–25 mg at bedtime
Migraine prophylaxis: Typically 10–50 mg daily
Dosage should be personalized and titrated by a physician
Best taken at bedtime due to sedative effect

Interaction:

Avoid with MAO inhibitors (serotonin syndrome risk)
Caution with CNS depressants, alcohol, and antihypertensives
May increase effects of anticholinergic and serotonergic drugs
May interact with thyroid medications and some SSRIs

Contraindications:

Recent myocardial infarction
Hypersensitivity to amitriptyline
Use with MAOIs within last 14 days
Glaucoma, urinary retention (due to anticholinergic effects)

Side Effects:

Common: Drowsiness, dry mouth, dizziness, constipation, weight gain
Less common: Blurred vision, urinary retention, sweating, orthostatic hypotension
Serious: Cardiac arrhythmias, seizures, suicidal ideation in young patients

Pregnancy & Lactation:
Use during pregnancy only if clearly needed. Amitriptyline is excreted in breast milk and may affect the infant; consult physician before use.
