Indications:
Prebalin 25 mg is indicated for the treatment of:
Neuropathic pain associated with diabetic peripheral neuropathy
Post-herpetic neuralgia
Fibromyalgia
Neuropathic pain associated with spinal cord injury
Adjunctive therapy for adult patients with partial-onset seizures
Pharmacology:
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system. This binding reduces the release of excitatory neurotransmitters, thereby decreasing neuronal excitability and providing relief from pain and seizures.
Dosage:
Neuropathic Pain & Post-herpetic Neuralgia:
Initial dose: 75 mg twice daily (150 mg/day)
Maintenance dose: 150–300 mg/day, divided into two or three doses
Fibromyalgia:
Initial dose: 75 mg twice daily (150 mg/day)
Maintenance dose: 300–450 mg/day, divided into two or three doses
Partial-onset Seizures:
Initial dose: 75 mg twice daily (150 mg/day)
Maintenance dose: 150–300 mg/day, divided into two or three doses
Administration:
Take orally with or without food.
Swallow the capsule whole with a glass of water.
Do not crush or chew the capsule.
Interaction:
Alcohol: May increase central nervous system (CNS) depressant effects.
Lorazepam: Concurrent use may enhance CNS depressant effects.
Clozapine: May increase blood levels of pregabalin.
Other CNS depressants: Concurrent use may increase the risk of CNS depression.
Contraindications:
Hypersensitivity to pregabalin or any component of the formulation.
Side Effects:
Common side effects include:
Dizziness
Somnolence
Dry mouth
Edema
Blurred vision
Weight gain
Abnormal thinking
Serious side effects may include:
Angioedema (swelling of the throat, head, and neck)
Respiratory depression
Suicidal thoughts or behavior
Pregnancy & Lactation:
Pregnancy: Category C. Use only if the potential benefit justifies the potential risk to the fetus.
Lactation: Pregabalin is excreted in breast milk. Use with caution during breastfeeding.
