Indications:
M-BEG-50-ER is indicated for the treatment of:
-
Neuropathic pain associated with diabetic peripheral neuropathy
-
Postherpetic neuralgia
-
Fibromyalgia
-
Generalized anxiety disorder
-
Adjunctive therapy in adults with partial-onset seizures
Pharmacology:
Pregabalin is a structural derivative of the inhibitory neurotransmitter GABA. It does not bind directly to GABA receptors but binds to the alpha-2-delta subunit of voltage-gated calcium channels in the CNS, reducing calcium influx and decreasing the release of excitatory neurotransmitters. The extended-release formulation allows once-daily dosing with sustained therapeutic effects.
Dosage:

Initial dose: 50 mg once daily

May be increased based on clinical response and tolerability

Maximum dose: 600 mg/day (divided or extended-release as appropriate)

Dose adjustment required for renal impairment
Administration:
Take orally once daily after an evening meal. Swallow the tablet whole; do not crush, split, or chew.
Interaction:

May enhance the sedative effects of CNS depressants like alcohol, opioids, and benzodiazepines
-
Concomitant use with other drugs affecting CNS should be done with caution
-
No significant interactions with other commonly used antiepileptics
Contraindications:

Known hypersensitivity to pregabalin

Severe renal impairment (dose adjustment mandatory)
Side Effects:
Common: Dizziness, drowsiness, dry mouth, peripheral edema, blurred vision
Less common: Weight gain, difficulty concentrating, ataxia
Serious: Suicidal thoughts, angioedema, hypersensitivity reactions
Pregnancy & Lactation:
Use in pregnancy only if the potential benefit justifies the risk to the fetus. Not recommended during lactation unless absolutely necessary and under medical supervision.
Precautions & Warnings:

Monitor for signs of suicidal ideation or behavior

Gradual tapering is required when discontinuing to avoid withdrawal symptoms

Use caution in patients with a history of substance abuse
