Indications:

THALIDON 25 MG is indicated for:

  • Multiple Myeloma (in combination with other anticancer agents such as dexamethasone)

  • Erythema Nodosum Leprosum (ENL), a painful inflammatory condition in leprosy
    It is also studied for use in various autoimmune and inflammatory disorders due to its immunomodulatory effects, but such use must be closely monitored.

Pharmacology:

Thalidomide exhibits immunomodulatory, anti-inflammatory, and anti-angiogenic properties. It suppresses tumor necrosis factor-alpha (TNF-α) and modifies cytokine release, which is useful in managing inflammatory diseases and cancers like multiple myeloma. It also inhibits angiogenesis (formation of new blood vessels), which helps limit tumor growth.

Dosage:

  • Multiple Myeloma: Usually initiated at 100–200 mg/day in combination with other drugs; however, THALIDON 25 MG may be used as a starting or adjusting dose.

  • ENL: Initial dose usually 100–300 mg/day, tapered based on response.

  • Dose must be individualized based on indication and patient condition.

Always follow your doctor’s dosage instructions precisely.

Administration:

  • Take orally, preferably at bedtime to minimize side effects like drowsiness.

  • Swallow the capsule whole with water.

  • Do not open, crush, or chew the capsule.

Interaction:

  • Concomitant use with sedatives, alcohol, or CNS depressants may increase drowsiness and dizziness.

  • May interact with drugs affecting the immune system or blood clotting.

  • Increases risk of neuropathy when used with neurotoxic drugs.

Always consult your physician before starting or stopping any medication.

Contraindications:

  • Absolute contraindication in pregnancy due to its severe teratogenic effects (causing birth defects).

  • Hypersensitivity to Thalidomide.

  • Women of childbearing potential not following strict contraceptive measures.

  • Severe neuropathy or uncontrolled cardiovascular conditions may also be limiting factors.

Side Effects:

Common side effects:

  • Drowsiness

  • Dizziness

  • Constipation

  • Peripheral neuropathy (tingling, numbness)

Serious side effects:

  • Blood clots (deep vein thrombosis, pulmonary embolism)

  • Severe skin reactions

  • Birth defects if used during pregnancy

Patients should be closely monitored for neurologic symptoms and thromboembolic events.

Pregnancy & Lactation:

  • Pregnancy: Strictly contraindicated. Thalidomide is highly teratogenic, even a single dose can cause severe birth defects. A Risk Management Program must be followed (e.g., REMS in some countries).

  • Lactation: Not recommended due to unknown excretion in breast milk and potential harm to the infant.
