Description
Tetraben 12.5 mg Tablet, manufactured by Incepta Pharmaceuticals Ltd., contains 12.5 mg of Tetrabenazine as its active ingredient. Tetrabenazine is a vesicular monoamine transporter 2 (VMAT2) inhibitor that works by depleting certain neurotransmitters—dopamine, serotonin, and norepinephrine—from nerve terminals in the brain. This mechanism is particularly effective in reducing hyperkinetic movements. Tetraben is primarily indicated for the treatment of chorea associated with Huntington's disease. It may also be used off-label for other hyperkinetic movement disorders such as tardive dyskinesia, senile chorea, and hemiballismus. The dosing regimen typically starts with 12.5 mg daily, gradually increasing to a maximum of 50 mg per day, depending on patient tolerance and response. The maximum single dose should not exceed 25 mg. Common side effects include sedation, fatigue, insomnia, depression, akathisia, anxiety, and nausea. Serious adverse reactions may include neuroleptic malignant syndrome and suicidal thoughts. Tetraben is contraindicated in patients with untreated depression, active suicidal ideation, hepatic impairment, or those taking monoamine oxidase inhibitors (MAOIs) or reserpine. Caution is advised when combining with strong CYP2D6 inhibitors, as dose adjustments may be necessary.
