Indications:
PG CR 165MG is indicated for the treatment of neuropathic pain associated with diabetic peripheral neuropathy, postherpetic neuralgia, as adjunctive therapy for partial onset seizures in adults, and generalized anxiety disorder. The controlled release formulation provides prolonged therapeutic effects.
Pharmacology:
Pregabalin, structurally related to GABA, does not bind directly to GABA receptors but modulates calcium channels by binding to the α2-δ subunit. This reduces excitatory neurotransmitter release (glutamate, substance P, norepinephrine), contributing to its analgesic, anxiolytic, and anticonvulsant properties. The CR formulation ensures gradual drug release over time.
Dosage:
The recommended starting dose is typically one 165 mg tablet once daily. The dose may be adjusted as per clinical response and tolerability under a physician’s supervision.
Administration:
Take the tablet orally once daily with or without food. Swallow whole—do not crush, split, or chew the tablet to preserve controlled release action.
Interaction:
May potentiate the effects of CNS depressants such as alcohol, opioids, and benzodiazepines. Monitor for increased sedation or dizziness. Caution with drugs affecting renal function.
Contraindications:
Hypersensitivity to Pregabalin or any excipient in the formulation.
Side Effects:
Drowsiness, dizziness, blurred vision, weight gain, dry mouth, fatigue, and peripheral edema are commonly reported. Controlled release form may reduce peak-related side effects.
Pregnancy & Lactation:
Pregabalin should only be used during pregnancy if the potential benefit justifies the potential risk. Caution is advised during lactation, as the drug is excreted in human milk.
Precautions & Warnings:
Avoid activities requiring mental alertness until the effects are known. Gradual withdrawal is recommended to avoid withdrawal symptoms. Renal function should be assessed prior to and during treatment.
