Indications:
Superitra (Itraconazole) is indicated for the treatment of systemic and superficial fungal infections including aspergillosis, blastomycosis, histoplasmosis, onychomycosis, and candidiasis of the oral and esophageal regions.
Pharmacology:
Itraconazole is a triazole antifungal agent that inhibits the synthesis of ergosterol, an essential component of fungal cell membranes, thereby disrupting cell membrane formation and function, leading to fungal cell death.
Dosage & Administration:
Typical dose for adults is 100-200 mg daily, depending on the infection. Capsules should be taken with food to enhance absorption. Dosage and duration vary by indication.
Interaction:
Itraconazole has significant interactions with drugs metabolized via the CYP3A4 pathway. Co-administration with drugs like simvastatin, quinidine, or cisapride is contraindicated due to risk of serious adverse effects.
Contraindications:
Contraindicated in patients with known hypersensitivity to itraconazole or any component of the formulation, and in patients with heart failure or ventricular dysfunction.
Side Effects:
Common side effects include nausea, abdominal pain, headache, rash, and elevated liver enzymes. Rare but serious reactions include hepatotoxicity and congestive heart failure.
Pregnancy & Lactation:
Itraconazole is contraindicated during pregnancy unless the potential benefit justifies the risk. Caution is advised during lactation as it may be excreted in human milk.
Precautions & Warnings:
Use with caution in patients with hepatic or renal impairment. Liver function should be monitored during prolonged therapy. Avoid grapefruit juice while on therapy.
