Indications:

SIDOPIN 5MG is indicated for the treatment of:

  • Essential hypertension

  • Chronic stable angina

  • Vasospastic angina (Prinzmetal's angina)

Pharmacology:

Amlodipine is a dihydropyridine calcium channel blocker that inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle. This action leads to vasodilation, reducing peripheral vascular resistance and lowering blood pressure. It also improves myocardial oxygen delivery in angina patients by dilating coronary arteries.

Dosage:

  • Adults:

    • Hypertension: Start with 5 mg once daily; may increase to a maximum of 10 mg daily based on patient response.

    • Angina: 5–10 mg once daily; most patients require 10 mg for optimal effect.

  • Elderly or patients with hepatic impairment:

    • Start with 2.5 mg once daily; adjust dosage based on clinical response.

Administration:

SIDOPIN 5MG can be taken orally with or without food. It is recommended to take the medication at the same time each day to maintain consistent blood levels.

Interaction:

  • May enhance the effects of other antihypertensive agents.

  • Caution when used with CYP3A4 inhibitors (e.g., ketoconazole, itraconazole) as they may increase plasma levels of amlodipine.

  • No significant interaction with digoxin or warfarin has been observed.

Contraindications:

  • Hypersensitivity to amlodipine or other dihydropyridine derivatives.

  • Severe hypotension.

  • Shock (including cardiogenic shock).

  • Obstruction of the outflow tract of the left ventricle (e.g., high-grade aortic stenosis).

  • Hemodynamically unstable heart failure after acute myocardial infarction.
