Indications:

  • Erectile dysfunction (ED) in adult men

Pharmacology:

Valenty 10mg Tablet contains vardenafil, a selective inhibitor of phosphodiesterase type 5 (PDE5). During sexual stimulation, nitric oxide is released in the corpus cavernosum of the penis, activating the enzyme guanylate cyclase, which increases levels of cyclic guanosine monophosphate (cGMP). cGMP causes smooth muscle relaxation and increased blood flow into the penis, leading to an erection. Vardenafil enhances this effect by inhibiting PDE5, which is responsible for the degradation of cGMP. It has no effect in the absence of sexual stimulation.

Dosage & Administration:

  • Initial dose: 10 mg orally once a day, taken approximately 60 minutes before sexual activity.

  • Adjustments: Based on efficacy and tolerability, the dose may be increased to 20 mg or decreased to 5 mg.

  • Maximum dose: 20 mg once a day.

  • Geriatric dose: For patients over 65 years, a starting dose of 5 mg is recommended.

  • Administration: Take with or without food.

Interaction:

  • Increased risk of hypotension when used with alpha-blockers.

  • May enhance the hypotensive effect of antihypertensive agents.

  • Caution when co-administered with other vasodilators.

  • Avoid use with nitrates or nitric oxide donors.

  • Increased serum levels when used with CYP3A4 inhibitors (e.g., ketoconazole, erythromycin).

Contraindications:

  • Concurrent use with nitrates or nitric oxide donors.

  • Hypersensitivity to vardenafil or any component of the tablet.

  • Severe hepatic impairment.

  • Severe renal impairment.

  • History of cardiovascular events such as stroke or heart attack within the last 6 months.

  • Uncontrolled high or low blood pressure.

  • Retinitis pigmentosa.

Side Effects:

  • Common: Headache, flushing, nasal congestion, dyspepsia, dizziness.

  • Serious: Priapism, sudden vision loss, hearing loss, chest pain, irregular heartbeat.

  • Others: Back pain, muscle aches, nausea.

Pregnancy & Lactation:

  • Pregnancy: Not indicated for use in women.

  • Lactation: Not recommended during breastfeeding.

Precautions & Warnings:

  • Use with caution in patients with cardiovascular diseases.

  • Monitor blood pressure regularly.

  • Discontinue use if priapism (prolonged erection) occurs.

  • Avoid alcohol consumption as it may increase the risk of side effects.

  • Not recommended for individuals under 18 years of age.
