Indications:
Lorat 10 mg is indicated for the symptomatic treatment of allergic rhinitis (seasonal and perennial) and chronic idiopathic urticaria (hives). It alleviates symptoms such as sneezing, runny nose, itching, and watery eyes.

Pharmacology:
Loratadine is a tricyclic antihistamine with selective peripheral H₁-receptor antagonistic activity. It is devoid of anticholinergic effects and has no sedative properties in the majority of the population. Loratadine works by blocking histamine receptors, thereby reducing allergy symptoms.

Dosage:

  • Adults and children over 12 years: 10 mg once daily.

  • Children 2 to 12 years:

    • Body weight over 30 kg: 10 mg once daily.

    • Body weight 30 kg or less: Not recommended; a more suitable dosage form should be used.

  • Children under 2 years: Not recommended due to insufficient safety and efficacy data.

Administration:
Take one tablet orally once daily, with or without food. Swallow the tablet whole with water.

Interaction:
Loratadine does not potentiate the effects of alcohol and has no significant interaction with other drugs. However, caution is advised when used with drugs that inhibit or are metabolized by hepatic cytochromes P450 3A4 and 2D6, as they may elevate plasma concentrations of loratadine.

Contraindications:
Contraindicated in patients with hypersensitivity to loratadine or any of the excipients in the formulation.

Side Effects:
Common side effects include headache and drowsiness. Serious side effects are rare but may include allergic reactions such as swelling of the face, lips, tongue, or throat, and difficulty breathing.

Pregnancy & Lactation:

  • Pregnancy: Category B. Should be used during pregnancy only if clearly needed.

  • Lactation: Caution should be exercised when administered to a nursing woman.
