Indications:

Pharmacology:
PADI D 400 Oral Solution contains cholecalciferol (vitamin D₃), which is metabolized in the liver to 25-hydroxyvitamin D₃ (calcidiol) and then in the kidneys to 1,25-dihydroxyvitamin D₃ (calcitriol). Calcitriol binds to vitamin D receptors, enhancing calcium and phosphate absorption in the intestines, promoting bone mineralization, and modulating immune system responses.
Dosage & Administration:
Infants (0–6 months): 0.5 ml twice daily (total 400 IU/day).
Children (>6 months): 1.0 ml twice daily (total 800 IU/day).
Pregnant Women: 400 IU daily; higher doses may be required if deficient (up to 2000 IU/day), as directed by a healthcare provider.
Administration:
Use the provided dropper for accurate dosing.
Administer directly into the mouth or mix with a small amount of food or drink.
Shake the bottle well before each use.
Interaction:
Medications like magnesium-containing antacids, digoxin, thiazide diuretics, cholestyramine, cholestipol, phenytoin, phenobarbital, orlistat, and mineral oil may interfere with vitamin D₃ absorption or metabolism.
Corticosteroids, such as prednisolone, may increase the body's requirement for vitamin D₃.
Contraindications:
Hypercalcemia or conditions associated with elevated calcium levels.
Known hypersensitivity to vitamin D₃ or any component of the formulation.
Evidence of vitamin D₃ toxicity.
Side Effects:
Rarely: Anorexia, nausea, vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, thirst, vertigo, and elevated calcium and phosphate levels in plasma and urine.
Pregnancy & Lactation:
Studies have shown safe use of doses up to 4000 IU during pregnancy. The recommended daily intake for pregnant women is 400 IU; however, in women who are considered to be vitamin D₃ deficient, a higher dose may be required.
Vitamin D₃ and its metabolites are excreted in breast milk. Overdose in infants induced by nursing mothers has not been observed; however, when prescribing additional vitamin D₃ to a breast-fed child, the practitioner should consider the dose of any additional vitamin D₃ given to the mother.
