Indications:
TOCIT 5 MG is indicated for prevention of organ rejection in patients receiving kidney, liver, or heart transplants. It may also be used in certain autoimmune diseases as prescribed.
Pharmacology:
Tacrolimus is a calcineurin inhibitor that suppresses T-lymphocyte activation by inhibiting calcineurin phosphatase activity. This reduces the immune response responsible for organ rejection.
Dosage:
Dosage varies according to transplant type and patient condition. Generally started at 0.1-0.2 mg/kg/day in divided doses, adjusted based on blood levels and clinical response.
Administration:
Orally, usually twice daily. Take consistently with or without food to maintain blood levels. Blood monitoring is required.
Interaction:
Tacrolimus interacts with drugs like CYP3A4 inhibitors (e.g., ketoconazole, erythromycin), nephrotoxic drugs, and immunosuppressants. Close monitoring is essential.
Contraindications:
Hypersensitivity to tacrolimus. Use caution in patients with infections or malignancies.
Side Effects:
Common side effects include nephrotoxicity, neurotoxicity, hypertension, hyperglycemia, tremors, and increased risk of infections.
Pregnancy & Lactation:
Use only if clearly needed during pregnancy. Tacrolimus is excreted in breast milk; breastfeeding is generally not recommended.
