Indications:
GLYMIN 500 MG is indicated for:

  • Management of type 2 diabetes mellitus

  • Used alone or in combination with other antidiabetic medications

  • Especially beneficial in overweight diabetic patients

  • Helps reduce the risk of diabetes-related complications

Pharmacology:
GLYMIN contains Metformin Hydrochloride, a biguanide class oral hypoglycemic agent. It works primarily by:

  • Decreasing hepatic glucose production

  • Reducing intestinal glucose absorption

  • Improving insulin sensitivity and peripheral glucose uptake

Unlike sulfonylureas, metformin does not stimulate insulin secretion, thereby reducing the risk of hypoglycemia.

Dosage & Administration:

  • Initial dose: 500 mg once or twice daily with meals

  • Maximum daily dose: Up to 2000–2500 mg (in divided doses)

  • Must be taken with food to minimize gastrointestinal side effects

  • Dose adjustments should be based on blood glucose monitoring and physician guidance

Interaction:

  • Alcohol may increase the risk of lactic acidosis

  • Cimetidine, furosemide, and nifedipine may affect metformin levels

  • May interact with contrast agents used in imaging; temporarily discontinue before procedures

Contraindications:

  • Renal impairment (eGFR < 30 mL/min/1.73 m²)

  • Acute or chronic metabolic acidosis, including diabetic ketoacidosis

  • Severe hepatic dysfunction

  • Conditions that predispose to hypoxia (e.g., heart failure, respiratory failure)

Side Effects:

  • Common: Nausea, diarrhea, abdominal pain, metallic taste

  • Rare but serious: Lactic acidosis (life-threatening)

  • Long-term use may reduce vitamin B12 absorption

Pregnancy & Lactation:

  • Category B: Generally considered safe during pregnancy under physician supervision

  • Use with caution during lactation

Precautions & Warnings:

  • Monitor renal function regularly

  • Avoid excessive alcohol intake

  • Stop temporarily before radiologic studies with contrast dye
