Indications:
PALZEN 0.5 MG is indicated for the prevention of:
Acute and delayed nausea and vomiting associated with highly or moderately emetogenic chemotherapy
Postoperative nausea and vomiting (PONV)
Pharmacology:
Palonosetron is a selective 5-HT3 receptor antagonist that prevents nausea and vomiting by blocking serotonin (5-HT) receptors in the chemoreceptor trigger zone (CTZ) and gastrointestinal tract. It has a long half-life (~40 hours), offering prolonged antiemetic protection compared to other drugs in its class.
Dosage & Administration:
For chemotherapy-induced nausea:
0.5 mg orally about 1 hour before chemotherapy
For postoperative nausea:
0.5 mg orally approximately 1 hour prior to anesthesia
Use as directed by a registered physician
Interaction:
Caution when combined with other serotonergic drugs (risk of serotonin syndrome)
No significant interaction with CYP450 enzyme inhibitors or inducers
Does not require dose adjustment with corticosteroids
Contraindications:
Hypersensitivity to Palonosetron or other 5-HT3 antagonists
Side Effects:
Common: Headache, constipation, dizziness
Less common: Fatigue, abdominal pain
Rare: Hypersensitivity reactions, serotonin syndrome (if used with other serotonergic agents)
Pregnancy & Lactation:
Pregnancy: Use only if clearly needed (Category B)
Lactation: Unknown if excreted in breast milk; caution advised
Precautions & Warnings:
Caution in patients with prolonged QT interval
Monitor for signs of serotonin syndrome when used with serotonergic drugs
Not for chronic use; use only before chemotherapy or surgery
