VORIFAST 40ML SUSPENSION is indicated for the treatment of severe and life-threatening fungal infections, including invasive aspergillosis, systemic candidiasis, and other rare fungal infections.
Pharmacology:
Voriconazole is a broad-spectrum antifungal agent that inhibits fungal cytochrome P450-dependent enzymes, reducing ergosterol synthesis. This action compromises the integrity of the fungal cell membrane, inhibiting fungal growth.
Dosage & Administration:
Adults:
Loading Dose: 6 mg/kg every 12 hours for the first 24 hours.
Maintenance Dose: 4 mg/kg every 12 hours, adjusted based on response.
Children (2-12 years): Weight-based dosing as advised by the physician.
Shake well before use. Administer at least one hour before or after meals.
Interaction:
Avoid co-administration with drugs like rifampin, carbamazepine, or long-acting barbiturates due to reduced effectiveness of Voriconazole.
Voriconazole may interact with medications like warfarin, cyclosporine, and phenytoin, requiring monitoring and dose adjustments.
Contraindications:
Hypersensitivity to Voriconazole or any components of the suspension.
Concurrent use with contraindicated drugs due to potential for severe interactions.
Side Effects:
Common: Visual disturbances, rash, nausea, and headache.
Serious: Hepatotoxicity, QT prolongation, and hypersensitivity reactions.
Pregnancy & Lactation:
Not recommended during pregnancy unless necessary due to potential risks to the fetus.
Use with caution during lactation, as the medication may be present in breast milk.
Precautions & Warnings:
Regular monitoring of liver and kidney function during therapy is recommended.
Avoid prolonged sunlight exposure due to photosensitivity risks.
Use cautiously in patients with pre-existing cardiac arrhythmias or electrolyte imbalances.
