Indications:
Amdocal 2.5mg is indicated for:
Mild to moderate essential hypertension
Chronic stable angina
Vasospastic angina (Prinzmetal’s angina)
First-line therapy in elderly or patients with hepatic impairment
Pharmacology:
Amlodipine is a dihydropyridine calcium channel blocker. It reduces vascular smooth muscle tone by blocking calcium influx, leading to vasodilation and decreased peripheral resistance. It also dilates coronary arteries, improving oxygen delivery in angina patients.
Dosage:
Adults: 2.5 mg to start, especially in elderly or hepatic impairment
May be increased gradually to 5–10 mg/day as needed
Children (6–17 years): Not generally recommended at this dose
Adjust dose based on clinical response
Administration:
Taken orally once daily
Can be taken with or without food
Try to take at the same time each day
Swallow whole with water
Interaction:
Increased effect when used with CYP3A4 inhibitors (e.g., ketoconazole)
May enhance the hypotensive effect of other blood pressure medicines
Monitor when combined with simvastatin or beta-blockers
Contraindications:
Known hypersensitivity to Amlodipine or other dihydropyridines
Severe hypotension
Cardiogenic shock
Severe aortic stenosis
Side Effects:
Common: Edema (especially ankle), dizziness, flushing, fatigue
Less common: Headache, palpitations, nausea
Rare: Liver enzyme elevation, gingival hyperplasia
Pregnancy & Lactation:
Pregnancy Category C
Use only if clearly needed and under physician supervision
Caution in breastfeeding; limited human data available
Precautions & Warnings:
Use with caution in patients with heart failure
Monitor BP closely when initiating treatment
Risk of hypotension, especially in volume-depleted individuals
May cause dizziness—advise patients to avoid driving initially
