Indications:
Amdocal Pro 2.5/5mg is indicated for:
-
Management of essential hypertension
-
Patients requiring dual therapy (ACE inhibitor + CCB)
-
High cardiovascular risk patients
-
Treatment of stable coronary artery disease
Pharmacology:
-
Perindopril (2.5mg): An ACE (Angiotensin-Converting Enzyme) inhibitor that reduces angiotensin-II production, causing vasodilation and decreased blood pressure.

Amlodipine (5mg): A dihydropyridine calcium channel blocker that relaxes vascular smooth muscle and lowers peripheral resistance.
Their combination leads to complementary blood pressure control and enhanced cardiovascular protection.
Dosage:

Standard dose: One tablet daily

Dose may be titrated based on therapeutic response

Not recommended for patients with severe renal impairment without specialist supervision
Administration:

Take orally, once daily
-
Preferably at a fixed time, with or without food
-
Swallow whole with water
-
Do not skip doses or stop abruptly
Interaction:
-
Risk of hyperkalemia with potassium-sparing diuretics or potassium supplements
-
Enhanced hypotensive effect with other antihypertensives
-
NSAIDs may reduce efficacy of Perindopril
-
Caution with lithium and certain diuretics
Contraindications:
-
Hypersensitivity to Perindopril, Amlodipine, or any ACE inhibitors

History of angioedema

Pregnancy (especially 2nd and 3rd trimesters)

Severe hypotension or aortic stenosis
-
Diabetic patients on aliskiren-containing drugs
Side Effects:
Common: Cough (from ACEI), dizziness, ankle swelling, headache
Less common: Taste disturbances, fatigue, abdominal pain
Rare: Angioedema, rash, increased liver enzymes
Pregnancy & Lactation:

Pregnancy Category D: Contraindicated due to risk of fetal harm

Not recommended during breastfeeding—consult physician
Precautions & Warnings:

Monitor renal function and electrolytes regularly

Use with caution in elderly and patients with renal artery stenosis
-
May cause dizziness—advise caution during driving or operating machinery
-
Not for use in combination with sacubitril/valsartan
