Indications:
DINOVO 500/20MG is indicated for the treatment of osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and for reducing the risk of NSAID-associated gastric ulcers in patients requiring long-term NSAID therapy.

Pharmacology:

  • Naproxen is a non-steroidal anti-inflammatory drug (NSAID) that inhibits prostaglandin synthesis through cyclooxygenase (COX) inhibition, reducing inflammation and pain.

  • Esomeprazole is a proton pump inhibitor (PPI) that reduces gastric acid secretion by blocking the H+/K+ ATPase in gastric parietal cells, thereby minimizing the risk of ulceration caused by prolonged NSAID use.

Dosage:

  • One tablet once or twice daily as prescribed by the physician.

  • Should be used at the lowest effective dose for the shortest duration necessary.

Administration:
Take orally at least 30 minutes before meals. Swallow the tablet whole; do not chew or crush.

Interaction:

  • Naproxen may interact with anticoagulants, diuretics, ACE inhibitors, and other NSAIDs.

  • Esomeprazole may interact with clopidogrel, digoxin, ketoconazole, and diazepam.

  • Avoid concurrent use with other ulcerogenic drugs unless advised by a physician.

Contraindications:

  • Hypersensitivity to Naproxen, Esomeprazole, or similar drugs.

  • History of gastrointestinal bleeding or perforation.

  • Severe hepatic or renal insufficiency.

  • Active peptic ulcer disease.

Side Effects:
May include abdominal pain, nausea, heartburn, headache, dizziness, constipation, or flatulence. Rare but serious effects: GI bleeding, kidney or liver dysfunction, allergic reactions.

Pregnancy & Lactation:
Not recommended during the third trimester of pregnancy. Should be used during pregnancy or lactation only if clearly necessary and under medical supervision.

Precautions & Warnings:

  • Caution in patients with a history of ulcers, cardiovascular disease, or kidney/liver disorders.

  • Avoid alcohol and smoking.

  • Monitor for gastrointestinal symptoms during prolonged use.

Therapeutic Class:
NSAID with gastroprotective agent (Fixed-dose combination)
