Indications:
Prosan 50mg is indicated for the treatment of major depression, anxiety, neuropathic pain, chronic tension headaches, migraine prevention, and insomnia associated with depression.
Pharmacology:
Prosan contains amitriptyline, a tricyclic antidepressant (TCA) that increases the levels of serotonin and norepinephrine in the brain by inhibiting their reuptake, thereby improving mood, relieving pain, and supporting sleep.
Dosage:
Adults: Usually starts at 25–50 mg per day, may be gradually increased to 75–150 mg/day depending on patient response.
Elderly and adolescents: Lower doses recommended.
Always follow physician’s instructions.
Administration:
Take orally once daily at bedtime or in divided doses with or without food. Do not crush or chew the tablet.
Interaction:
May interact with MAO inhibitors, SSRIs, anticholinergics, CNS depressants, antihypertensives, and alcohol. Risk of serotonin syndrome with serotonergic agents.
Contraindications:
Not recommended in patients with hypersensitivity to amitriptyline, recent heart attack, cardiac arrhythmia, or taking MAO inhibitors.
Side Effects:
Common: Drowsiness, dry mouth, dizziness, weight gain, constipation.
Rare but serious: Irregular heartbeat, seizures, serotonin syndrome, suicidal thoughts in young patients.
Pregnancy & Lactation:
Should be used during pregnancy only if clearly needed. Caution is required during breastfeeding, as the drug may pass into breast milk.
Precautions & Warnings:
Monitor patients with cardiovascular disease, epilepsy, liver impairment, or bipolar disorder. Avoid abrupt discontinuation and alcohol.
